On my NP435 there are three 3/4" holes on the flat on the front of the top cover, two are plugged one is open is this for a vent tube or should i plug this as well so as not to fill the trans with water?

What should i plug it with if it is to be plugged?

I'm sure Elwinil will know this.

One of them should have the backup light switch in it. There is a vent on the transmission too but not sure where it is located.

If you are talking about the holes that are on the vertical flat at one end of the cover (mine are on the rear of the cover) then those are the holes for the shift rails to be inserted and they should all be plugged.

yup those are the holes

How do you plug them? where can i find some new plugs?

Any parts store should carry 3/4" freeze plugs.  That should work fine.  The Mopar Parts Manual lists the part number but the description simply says PLUG, Rail Hole Expansion 3/4" so that is the correct size.
